Process Development Scientist ( Downstream, Bioprocess, Viral Vectors, AAV) 305174

RESPONSIBILITIES:

• 
  
• Bioprocess;- Downstream Process Development, process optimization for manufacturing viral vectors
  
• Vector Engineering/ Viral Vectors. Support vector core Vector Engineering processes, Process Development
  
• Support vector core vector engineering processes, process development, and process optimization
  
• Process Development, process optimization, Bioprocess
  
• Upstream/ Downstream Process Development, process optimization for manufacturing viral vectors
  
• Bioprocess development of AAV vectors focusing, upstream cultivation and harvest conditions to optimize yield
  
• Working with AAV viral particles, fermentation, plasmid recovery, purification and formulation, sterilization, and harvesting process steps
  
• Apply research/ industry knowledge of Lentivirus (LV) and or Adeno Associated Virus (AAV)
  
• Cell Line maintenance, Cell Line development, Media development; as cell are transfected with plasmids to generate viral vectors, which are efficiently harvested
  
• Creating and maintaining stable cell lines
  
• Troubleshooting and optimizing stepwise.
  
• Plasmid Preparation, Buffer, Media Preparation, Aseptic Processing, growth & maintenance of cell lines.
  
• Handling Bioreactors, reagent/media prep, sterile manufacturing, upstream/downstream processing
  
• Transfection, generate viral vectors, and efficient harvesting
  
• Troubleshooting and optimizing processes stepwise.
  
• Apply skills in cGMP and gene therapy, vector purification, process scale-up, and sterile manufacturing
  
• Bioprocess development of AAV vectors focusing, upstream cultivation and harvest conditions to optimize yield
  
• Managing suspension cultures, downstream purification recoveries, and vector potency
  
• Overall support of upstream process development for novel gene therapy vector.

  REQUIREMENTS:
  
  
  • 
    
  • Bachelor's (BS) or Master's (MS) in Molecular Biology, Virology, Biology, Biochemistry or related
    
  • Minimum of a 4-8 yrs. + experience in the Biotech, Cell & Gene Therapy (CGT) industry working with viral vectors
    
  • In depth knowledge of both upstream processing and or downstream processing
    
  • Knowledge of protein purification, mammalian cell culture, virology, viral validation, Preformulation
    
  • Formulation, ultracentrifugation, flocculation, scale-able chromatography and tangential flow filtration (TFF).
    
  • Process Development (PD), manufacturing of viral vectors; transfection, transcription, translation, AAV vector
    
  • in vivo genetic modification, Plasmid DNA Extraction
    
  • 4-8 yrs. + experience handling bioreactors, media development, and process development/ bioprocess refinement
    
  • 4-8 yrs. + experience in protein and gene therapy, vector purification, process scale-up, and sterile manufacturing
    
  • Upstream/Downstream and the equipment and tools used in that process including; bioreactors, Vi-CELL, S.U.M.'s, sterile tubing welders, filters, balances, microscopes, pipettes, CellSTACK's, HYPERstack's Clipsters, DeltaV, BSC's, pH Meters, Osmometers, Autoclaves etc.
    
  • Sterile manufacturing, cGMP, cleanroom, Upstream / Downstream Process Development and GMP compliance.
    
  • UPSTREAM - 20L-200L bioreactors, cell expansion, transient transfection, viral production and clarification
    
  • DOWNSTREAM -Recovery and purification of viral vectors and removal of contaminants. Nuclease digestion, Purification, Sterile Filtration and Fill
    
  • ANALYTICS - PAT, feed rates, pH control, Potency measurement, Bioburden, Mycoplasma, Endotoxin
    
  • cGMP, sterility, fermentation, mammalian cell culture, bioprocess optimization, purification, including clarification by filtration/ centrifugation; chromatography, ion exchange, sizing, multi-modal and other resins
    
  • Tangential Flow Filtration (TFF) or Ultracentrifugation/ Diafiltration (UF/DF), Size Exclusion Chromatography.
